Mitochondrial Peptide Therapy: MOTS-c

Restore Cellular Energy, Metabolism, and Performance at the Source

Declining mitochondrial function is one of the recognized hallmarks of biological aging.1

Mitochondria are the energy centers inside nearly every cell. They control how efficiently your body converts food into fuel, how well you respond to exercise, how quickly you recover, and how resilient you feel under stress. Loss of mitochondrial function has been linked to fatigue, exercise intolerance and muscle weakness, and to the disability that develops with age.2

MOTS-c is a mitochondrial-derived peptide that acts as a metabolic signal between mitochondria and the rest of the cell.3

EnergizeMe pathway › MOTS-c Also in EnergizeMe: SS-31
What Is MOTS-c?

What Is MOTS-c?

MOTS-c is a naturally occurring peptide your body already produces.4 In laboratory and animal studies, it acts as a metabolic messenger that regulates insulin sensitivity and metabolic homeostasis, in part through AMPK activation.4

Circulating MOTS-c levels have been reported to be lower in older people and in people with type 2 diabetes. Whether lower MOTS-c causes slower metabolism or reduced exercise response in humans has not yet been established.56

In older mice, MOTS-c treatment increased physical capacity; comparable benefits have not yet been shown in human clinical trials. However, anecdotally, positive patient outcomes are noted.78

Benefits of MOTS-c Peptide Therapy

Patients enrolled in our EnergizeMe mitochondrial optimization program may experience:

Who Is MOTS-c Best For?

Who Is MOTS-c Best For?

MOTS-c may be right for you if:

  • You feel chronically fatigued despite getting enough sleep
  • You struggle with weight that resists diet and exercise changes
  • Your workout performance or recovery has noticeably declined
  • You experience post-meal energy crashes or blood sugar instability
  • You feel “metabolically stuck” despite doing the right things
  • You’re dealing with brain fog, low motivation, or difficulty concentrating
  • You want a longevity-focused, foundational approach to aging well

This program is often combined with structured nutrition, movement guidance, and lifestyle optimization for the best results. Dr. Varano or a member of his Medical Provider Team will review your full picture before recommending a protocol.

Month 4 — Add SS-31 On Your Terms:

When you reach Month 4 of your cycle, you have the option to add a 16-day SS-31 protocol. SS-31 binds cardiolipin in the inner mitochondrial membrane; whether adding it to MOTS-c improves results has not yet been studied.

SS-31 Add-On in Month 4 (pricing discussed at your assessment)

$275/month

The preferred stacking protocol — MOTS-c and SS-31 working together as a structured, fully managed mitochondrial optimization program.

Why stack MOTS-c with SS-31?

MOTS-c is a mitochondrial-derived signaling peptide, while SS-31 binds cardiolipin in the inner mitochondrial membrane. They act by different mechanisms, but the combination has not yet been studied in clinical trials and a greater benefit than either alone has not yet been shown.

What is MOTS-c peptide therapy?

MOTS-c is a naturally occurring mitochondrial-derived peptide that regulates how the body produces and uses energy. In animal studies, MOTS-c treatment improved insulin sensitivity and physical performance. These effects have not yet been confirmed in human clinical trials. Anecdotally, though, patients do report positive outcomes.47

MOTS-c is not an appetite suppressant or a stimulant. It is a signaling peptide whose metabolic effects have so far been shown mainly in cell and animal studies. Weight or body-composition changes with MOTS-c have not yet been demonstrated in human clinical trials, and it is not a weight-loss drug.48

MOTS-c may be considered for patients experiencing fatigue, metabolic slowdown, exercise resistance, insulin sensitivity issues, or age-related declines in energy and recovery, although its benefit in these groups has not yet been established in human trials. However, anecdotally, positive patient outcomes are noted. It is well-suited for anyone seeking a longevity-focused, foundational approach to metabolic health.

There are no human clinical trial data on how quickly MOTS-c works, and responses vary between individuals. Metabolic and body composition changes typically develop over time. Results vary based on baseline health, activity level, nutrition, and adherence to your overall program.

SS-31 (Elamipretide) is a mitochondria-targeted peptide that binds cardiolipin in the inner mitochondrial membrane. MOTS-c is a mitochondrial-derived signaling peptide. The two act by different mechanisms; their combination has not yet been studied in clinical trials.11 SS-31 is part of the EnergizeMe Suite.

SS-31 is administered as a 16-day treatment protocol and is available up to 3 times per year. It can be added to your Standard EnergizeMe plan in Month 4 (pricing discussed at your assessment), or included automatically in every cycle under the Premium EnergizeMe Stack at $275/month.

Growth hormone secretagogues like Sermorelin and CJC-1295 + Ipamorelin work by stimulating the pituitary gland to produce more growth hormone — supporting muscle, recovery, body composition, and sleep quality. MOTS-c works differently: it is a mitochondrial-encoded signaling peptide that improved insulin sensitivity and metabolic homeostasis in animal studies.4 Think of MOTS-c as restoring your cellular power plants; growth hormone peptides as boosting your body’s master recovery signal. Some patients benefit from both, depending on their goals.

Human safety data for MOTS-c are very limited: the FDA states it has not identified human exposure data for drug products containing MOTS-c and lacks important information about its safety.128 MOTS-c has not been studied in pregnancy or breastfeeding and should not be used during either. Dr. Varano will review your full medical history and any medications before recommending this therapy.

Results are enhanced when MOTS-c therapy is paired with regular physical activity, balanced nutrition, adequate sleep, and lifestyle optimization. Your program includes guidance to support these areas.

Yes. MOTS-c is offered exclusively within the YoungerMeMD membership model to ensure proper medical oversight, continuous monitoring, and long-term optimization. Membership is required for all peptide therapy programs.
